SPRI-Based Cleanup & Library Prep

High Molecular Weight Size Selection.

Streamline your workflow. Cleanup and library prep you can trust.

Library preparation and PCR-based workflows demand consistent, high-purity nucleic acids free of primers, adapter dimers, enzymes, and salts. Yet traditional column-based cleanup and outdated SPRI bead solutions slow down throughput, limit automation, and compromise reproducibility.

Introducing MAGic™SPRI HMW, the solid-phase reversible immobilisation (SPRI) bead solution from MAGic Bioprocessing, engineered for the evolving needs of library preparation workflows. Its superior magnetic response enables faster bead resuspension just a few pipette cycles, more efficient magnetic separation, and complete bead pelleting without sticking to pipette tips or well surfaces, eliminating sample loss and reducing hands-on time at every step. Where standard SPRI beads slow you down and compromise your cleanup, MAGicSPRI HMW sets a new standard for SPRI-based purification: faster, cleaner, and effortlessly consistent from tube to 96-well plate.

Addressing the Challenges of Library Preparation and Nucleic Acid Cleanup

The quality of a sequencing run is only as good as the library that feeds it. As sequencing workflows have become central to genomics, oncology, metagenomics, and translational research, the demands on nucleic acid cleanup and precise fragment size selection have grown significantly. Researchers and production labs alike face a set of recurring challenges:

Inconsistent size selection

Standard SPRI beads often deliver variable base pair cut-offs, leading to libraries with broad size distributions that reduce efficiency.

Unwanted contaminant carryover

Incomplete removal of primer dimers, adapter dimers and unincorporated dNTPs reduces sequencing data quality, inflates error rates, and causes adapter over-representation in downstream data analysis.

Slow magnetic separation

Low-magnetisation beads require minutes to pellet, a bottleneck in high-throughput 96-well plate cleanup workflows and automated liquid-handling systems, where seconds matter.

There is a clear need for a faster, more precise, and automation-ready SPRI bead solution that can deliver narrow fragment size selection consistently, across every sample.

MAGic™SPRI HMW – Where SPRI Meets Magic

MAGic™SPRI HMW are super-paramagnetic solid beads based on solid-phase reversible immobilisation (SPRI) technology, optimised for high-efficiency nucleic acid binding. Designed for both cleanup and library preparation workflows, they deliver exceptional nucleic acid purification and precise size selection within a broad range of 100 bp up to 5 kb, making them ideal for high molecular weight (HMW) isolations.

Validated Performance

Precise size selection

Size-selective performance across seven bead ratios (0.3x–1.8x) retaining fragments from 100 bp up to 5 kb. Single-sided and double-sided size selection validated on E-Gel.

High reproducibility

Four replicates of an RNA sequencing library pool with the average selected fragment size was 582.5 nucleotides with a standard deviation of just 3.5 nucleotides, a coefficient of variation of 0.6%, demonstrating excellent consistency across automated workflows.

Long read optimised

A pooled 16S rRNA gene amplicon library purified with MAGic™SPRI HMW TapeStation analysis confirmed excellent library profiles, and subsequent Oxford Nanopore Technologies (ONT) sequencing delivered identical outputs between samples.

Fast handling, easy resuspension

MAGic™SPRI HMW beads separate within seconds on a standard magnet. They do not aggregate, resuspend uniformly with minimal mixing steps, and do not adhere to pipette tips or well surfaces, making them ideal for automated liquid-handling platforms and 96-well plate formats.

MAGic™SPRI HMW delivered sequencing outputs identical to the Market Leader in an Oxford Nanopore 16S amplicon sequencing workflow, at a fraction of the cost.

Why MAGic™SPRI HMW?

Conventional SPRI bead cleanup solutions were not designed for today’s high-throughput, automation-driven library preparation workflows. MAGic™SPRI HMW is engineered to overcome their limitations:

  • Speed: Magnetic separation within seconds, not minutes for standard SPRI beads, cutting hands-on time and cost per sample.
  • Precision: Validated 0.6% CV in fragment size, across automated replicates.
  • Easy handling: Superior magnetisation means beads pellet completely, resuspend in just a few pipette cycles, and don’t stick to tips or well surfaces. Less bead loss, fewer failed runs, lower cost per sample.
  • Drop-in compatibility: Fully interchangeable with standard SPRI protocols.
  • Full-range size selection: Fragment enrichment from 100 bp all the way up to 5 kb, covering everything up to high molecular weight DNA isolation in a single bead solution.
  • Cost-effective nucleic acid purification: Faster separation, fewer pipetting steps, and no bead loss combine to reduce reagent waste and operator time, making MAGicSPRI™ HMW the most cost-effective SPRI technology choice for high-throughput labs.

MAGic™SPRI HMW, the cost-effective SPRI bead solution that brings speed and precision to every cleanup and library preparation workflow.
